Yes. Read the rules.

Can a character use a defense card against a special card, such as Boba Thermal Detonator card?

No. Read the rules.

If two characters are next to each other diagonally, are they considered adjacent for attack purposes?

Yes! Rules! Read!

Can characters can fire over the blue mist in the Emperor's Throne Room even if both they and their target are on the ground level?

Yes. Raised obstacles block ranged attacks, lowered obstacles do not.

Note: The simple convention of this rule is often broken or ignored in custom maps.

Can you play a card that says "does not count as an action" after you have alredy taken two actions on a turn?

No, your turn ends immediately upon taking your last allotted action. You must play such a card before you take your final action on a turn. If you have no allotted actions, you cannot play a "non-action" card.

When your minor character(s) is (are) dead, you can play their cards to heal damage to your major character. But what happens to cards which don't belong to the minor character, but are still worthless without the minor characters? For example, the Emperor's Royal Command; card is worthless if both guards are dead.

Such cards are dead weight. (Royal Command is dead weight anyway, am I right!?)

If a player cycles their deck twice, who wins?

The rules and the designers have left us with an Answerless FAQ. I recommend you keep playing until someone is eliminated.

Ulitmately, you have to decide for yourself.
